Overview

NADH:Ubiquinone Oxidoreductase Subunit A7 (NDUFA7) is a nuclear-encoded accessory subunit of the mitochondrial membrane respiratory chain NADH dehydrogenase (Complex I). Complex I is the first and largest enzyme of the mitochondrial electron transport chain, responsible for transferring electrons from NADH to ubiquinone and pumping protons across the inner mitochondrial membrane to drive ATP synthesis (UniProt O95182). The NDUFA7 mRNA encodes a protein that is essential for the proper assembly and structural integrity of the Complex I holoenzyme (PMID: 29335508). Mutations in the NDUFA7 gene or alterations in its mRNA expression are associated with Mitochondrial Complex I deficiency, which can lead to severe metabolic and neurological disorders such as Leigh syndrome (PMID: 30245028). In the context of drug development, NDUFA7 is considered a component of the Complex I target for small-molecule inhibitors like metformin or IACS-010759, which are investigated for their ability to modulate metabolic pathways in cancer and metabolic diseases (NCBI Gene 4701). Furthermore, NDUFA7 mRNA levels serve as a critical biomarker for assessing mitochondrial function and the efficacy of therapies aimed at restoring oxidative phosphorylation.
