NADPH-cytochrome P450 oxidoreductase (POR), also known as CPR or CYPOR, is a membrane-bound flavoprotein enzyme that transfers electrons from NADPH to all microsomal cytochromes P450 and other heme-containing proteins. This electron transfer is essential for drug metabolism, steroid hormone biosynthesis, cholesterol and fatty acid homeostasis, and the detoxification of xenobiotics. POR consists of an FMN-binding domain, a connecting domain, an FAD-binding domain, and an NADPH-binding domain. It is anchored to the endoplasmic reticulum membrane by a hydrophobic N-terminal transmembrane segment. POR catalyzes sequential electron transfers from NADPH to FAD to FMN to the final acceptor (e.g., cytochrome P450).
