NADPH oxidase 3 is a member of the NOX family of oxidoreductase enzymes that generate superoxide and other reactive oxygen species (ROS) by transporting electrons from NADPH across biological membranes. NOX3 is predominantly expressed in the inner ear, where it is critical for the formation of otoconia/otoliths necessary for the perception of gravity and normal vestibular function. Like other NOX family members, NOX3-derived ROS play roles in cellular signaling, regulation of gene expression, and cell differentiation. Mutations or dysregulation in NOX3 are linked to specific clinical phenotypes, such as drug-induced hearing loss and disorders of immune defense. While the NOX family is implicated in a range of ROS-driven diseases, selective pharmacological targeting of NOX3 specifically is limited, and direct clinical biomarkers or therapies are not yet established.
