NADPH oxidase 4 (NOX4) pre-mRNA is the precursor transcript of the NOX4 enzyme, a member of the NADPH oxidase family that uniquely produces hydrogen peroxide constitutively. Unlike other NOX isoforms that require complex assembly for activation, NOX4 activity is primarily regulated at the level of gene expression and pre-mRNA processing, making its transcript a critical control point for cellular redox homeostasis. Pathological upregulation of NOX4 is a hallmark of several fibroproliferative and inflammatory conditions, including idiopathic pulmonary fibrosis and diabetic nephropathy, where excessive ROS production promotes myofibroblast activation and extracellular matrix deposition. Therapeutic interventions targeting NOX4 pre-mRNA utilize antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs) to reduce the pool of mature mRNA through RNase H-mediated cleavage or RNA interference. These RNA-targeted approaches offer a highly specific means of silencing NOX4 expression, potentially overcoming the selectivity issues often encountered with small-molecule inhibitors of the NOX protein family.
