Overview

NADPH oxidase activator 1 (NOXA1) is a cytosolic regulatory protein that acts as a key activator of the NOX1 isoform of NADPH oxidase, an enzyme complex involved in the controlled production of reactive oxygen species (ROS) such as superoxide[1][3][5]. NOXA1 is functionally analogous to p67phox (NCF2) in phagocytic NOX2 systems but is specific to non-phagocytic NOX1 complexes, where it partners with NADPH oxidase organizer 1 (NOXO1) to facilitate the assembly and activation of the NOX1 enzyme at the cell membrane[1][5][8]. This activation is crucial for ROS-mediated cellular signaling, innate immune defense, and processes such as cell proliferation and tissue remodeling[2][5]. Dysregulation or altered expression of NOXA1 has been associated with inflammatory diseases, cancer (especially colon cancer, where it was described as NY-CO-31), and other pathological conditions involving redox imbalance[3][5][8]. Drugs targeting NADPH oxidase activity are in experimental use, but no clinical agents selectively targeting NOXA1 are currently available[6].
