Overview

The NADPH oxidase complex is a membrane-bound, multi-protein enzyme complex responsible for generating superoxide, a reactive oxygen species essential for immune cell microbial killing and various redox-dependent cellular functions[1][3][4][5][6][7]. It consists of a catalytic core (NOX isoforms such as NOX2/gp91^phox^) complexed with regulatory subunits (including p22^phox^, p47^phox^, p67^phox^, and Rac GTPase) whose assembly at the membrane is tightly regulated and necessary for enzymatic activity[1][2][3][7]. Impaired complex assembly/function underlies disorders like chronic granulomatous disease, while excessive or dysregulated ROS production contributes to cardiovascular, inflammatory, and neurodegenerative diseases[1][7]. Restoration of NADPH oxidase complex assembly/function is thus both a concept for treating immunodeficiency and a target for inhibiting pathological ROS production in non-infectious diseases[1][4][7].
