Overview

NADPH Oxidase Organizer 1 (NOXO1) is a cytosolic protein that functions as an organizer subunit for NOX1 and NOX3 NADPH oxidases. It is essential for their assembly and activation, resulting in the production of reactive oxygen species (ROS), such as superoxide. NOXO1 contains a PX domain for phospholipid binding and two SH3 domains for protein-protein interactions. It plays a role in otoconia biogenesis and modulates Notch signaling via ADAM17 regulation, impacting endothelial cell behavior and angiogenesis. Dysregulation of NOXO1 is implicated in oxidative stress-related diseases, diabetes-related kidney damage, and vascular dysfunction.
