NALCN channel auxiliary factor 1 (NALF1), also known as FAM155A, is a transmembrane protein that functions as an essential auxiliary component of the NALCN sodium leak channel complex[1][3][6]. It facilitates proper folding, assembly, and membrane localization of the NALCN channel, which is responsible for the basal sodium permeability of neurons and plays a critical role in maintaining resting membrane potential and regulating excitability in the central nervous system[2][3][6]. NALF1 is predicted to contribute to stretch-activated, cation-selective channel activity and is involved in calcium ion import across the plasma membrane[1][3]. Mutations or dysfunction in the NALCN complex, including its auxiliary factors such as NALF1, have been implicated in severe neurodevelopmental syndromes, although NALF1 itself is not currently an established therapeutic target and no drugs are known to act on it directly[3][2][6].
