Nanog homeobox pseudogene 1 (NANOGP1) is a duplicated pseudogene highly homologous to the transcription factor NANOG, located at chromosome 12p13.31. While traditionally annotated as a non-protein-coding pseudogene, recent work has shown expression of a NANOGP1-derived protein in naïve human pluripotent stem cells, with overlapping and distinct roles compared to NANOG. Unlike processed pseudogenes, NANOGP1 arose by tandem duplication and retains regulatory elements. It is expressed primarily in naïve hPSCs and during early embryogenesis. Functional studies suggest partial conservation of pluripotency-related activities, including potential autorepressive capacity and the ability to contribute to reprogramming under minimal conditions. However, it is not required to maintain pluripotency. No evidence exists that NANOGP1 is a validated or druggable therapeutic target, nor is it currently associated with disease or clinical biomarkers. There is some confusion in the literature and public databases, as "NANOGNBP1" is not an official symbol for any human gene or pseudogene. "NANOGNB" refers to a different, highly divergent NANOG paralogue unrelated to NANOGP1. Key clarifications: - "NANOGNB pseudogene 1" and "NANOGNBP1" are non-standard and potentially incorrect aliases; the standardized name is "Nanog homeobox pseudogene 1" (NANOGP1). - NANOGP1 is not established as a therapeutic target and should be considered a pseudogene with possible protein-coding function in human pluripotent cells. - No drugs or clinical interventions are known to act on this gene. - While recent work challenges its pseudogene status, there remains ambiguity; thus, use with caution for drug-target or biomarker studies.
