Overview

Nanos homolog 3 (NANOS3) is an evolutionarily conserved RNA-binding protein featuring two unique C2HC-type zinc finger motifs that are indispensable for its function in germ cell development and maintenance[1][2][4][5]. NANOS3 is expressed in human gonads, localizing predominantly to the nuclei of germ cells during spermatogenesis and oogenesis[2][4]. Its primary biological role is to maintain germ cell populations by regulating the expression of genes required for pluripotency and proper meiotic initiation; NANOS3 acts as an anti-apoptotic factor in primordial germ cells, protecting them during critical developmental windows[2][3][4]. Loss or reduction of NANOS3 function leads to decreased germ cell numbers and is associated with disorders such as primary ovarian insufficiency and infertility, likely through impaired anti-apoptotic activity and disturbed gene regulation[4]. NANOS3 functions by binding RNA—without strict sequence specificity—and collaborates with other translational regulators such as Pumilio to repress mRNA translation during early germ cell development[1][5]. There are currently no drugs or established therapeutic mechanisms directly targeting NANOS3.
