Nasal mucous membrane receptors is a collective term referring to a heterogeneous group of receptor proteins located within the respiratory and olfactory epithelia of the nasal cavity. This group includes alpha-adrenergic receptors (alpha-1 and alpha-2) that regulate vascular tone and mucosal thickness, and histamine H1 receptors that mediate allergic responses like sneezing and itching (StatPearls: Oxymetazoline, 2023). The mucosa also contains glucocorticoid receptors, which are targeted by intranasal steroids to manage chronic inflammation in allergic rhinitis and nasal polyposis, and muscarinic receptors that control mucus production by seromucous glands. Furthermore, the specialized olfactory epithelium houses a large family of olfactory receptors (G protein-coupled receptors) responsible for the detection of odorants and the sense of smell (Nature: Olfactory Receptors, 2014). Because this designation refers to an anatomical collection of various molecular targets rather than a single specific protein, it is considered a broad functional classification rather than a distinct therapeutic target in molecular biology. Local delivery of drugs to these receptors allows for high therapeutic concentrations at the site of disease with minimal systemic side effects.
Agonism of alpha-1 and alpha-2 adrenergic receptors induces vasoconstriction of nasal mucosal blood vessels to reduce swelling; antagonism of Histamine H1 receptors inhibits allergic inflammation and symptoms; activation of glucocorticoid receptors provides broad anti-inflammatory effects; antagonism of muscarinic receptors reduces glandular secretions (rhinorrhea).
