Natriuretic peptide receptors (NPRs) are a family of cell surface receptors that mediate the physiological effects of natriuretic peptides—hormones involved in cardiovascular and renal homeostasis. There are three main subtypes: NPR-A, NPR-B, and NPR-C. NPR-A and NPR-B are guanylyl cyclases that produce cGMP, while NPR-C primarily functions as a clearance receptor. They are involved in vasodilation, natriuresis, diuresis, and skeletal growth regulation, making them therapeutic targets for cardiovascular and skeletal disorders.
Agonist binding leads to cGMP production and downstream signaling (NPR-A/B). NPR-C mediates internalization and degradation of natriuretic peptides.
