Overview

Natural cytotoxicity receptors (NCRs) are a family of activating receptors expressed primarily on natural killer (NK) cells, including NKp30, NKp44, NKp46, and NKp80, that trigger NK cell cytotoxicity and cytokine secretion (e.g., IFNγ) against tumor cells, virus-infected cells, and pathogens via ITAM-containing adapters like DAP12 and signaling through Syk/Zap70 kinases. They recognize diverse ligands such as B7-H6, heparan sulfate proteoglycans, and pathogen-derived molecules, with splice variants enabling activating or inhibitory functions depending on context, and play key roles in tumor surveillance, viral control, and immune regulation in tissues like decidua.
