Overview

Natural cytotoxicity triggering receptor 1 (NKp46) ligands on abnormal or tumor cells are a heterogeneous group of molecules that trigger Natural Killer (NK) cell activation upon binding to the NKp46 receptor. These ligands include endogenous proteins such as surface-exposed calreticulin, vimentin, and specific heparan sulfate proteoglycans (HSPGs), as well as exogenous viral hemagglutinins [1][2]. The interaction is a key component of the immune system's ability to recognize and eliminate stressed, infected, or malignant cells through the induction of cytotoxicity and cytokine production [3]. In the context of oncology, tumors often downregulate these ligands or shed them in soluble forms to evade immune detection, a process that correlates with poor prognosis [4]. Therapeutic development focuses on multi-specific NK cell engagers (NKCEs) that bypass natural ligand requirements by directly linking the NKp46 receptor to tumor-associated antigens [5]. Additionally, certain treatments like anthracycline chemotherapy can induce the translocation of ligands like calreticulin to the cell surface, promoting immunogenic cell death [6].
