Natural killer (NK) cell chemokine receptors are a group of G protein-coupled receptors (GPCRs) expressed on the surface of NK cells that mediate their migration toward chemokine gradients within the tumor microenvironment (TME). Key receptors include CXCR3, CX3CR1, CCR2, CCR5, and CXCR4, which respond to specific ligands like CXCL9, CXCL10, and CCL5 produced by tumor or stromal cells. In the context of oncology, these receptors are critical for the successful infiltration of NK cells into solid tumors, where they exert potent anti-tumor cytotoxic effects. Therapeutic strategies often focus on enhancing the expression of these receptors on NK cells (e.g., via CAR-NK engineering) or modulating the TME chemokine profile to improve the recruitment and retention of these effector cells. Conversely, some chemokine pathways are targeted for inhibition if they contribute to immune evasion or the recruitment of regulatory cell types that suppress NK cell activity.
