Overview

Natural killer (NK) cell cytokine receptors are a diverse group of cell surface proteins expressed on NK cells that mediate signaling in response to various cytokines, including IL-2, IL-12, IL-15, IL-18, and IL-21. These receptors, such as the common gamma chain (CD132) and the IL-2/IL-15 receptor beta subunit (CD122), are essential for the development, survival, and functional activation of NK cells. Upon ligand binding, these receptors typically activate the JAK/STAT signaling pathway, which triggers the production of effector molecules like interferon-gamma (IFN-γ) and cytotoxic granules (perforin and granzymes) necessary for the elimination of tumor and virally infected cells. In therapeutic development, these receptors are primary targets for immunotherapies aimed at boosting NK cell activity, with drugs like IL-15 superagonists (e.g., N-803) and IL-2 variants (e.g., aldesleukin) being used to enhance anti-tumor responses. However, the use of these agents is often limited by systemic toxicities such as cytokine release syndrome and vascular leak syndrome, which result from the broad activation of immune pathways. The expression levels of specific subunits, such as CD122 and CD132, often serve as biomarkers for NK cell maturity and responsiveness to cytokine-based therapies.
