Natural killer cell cytotoxicity receptor 3 ligand 1 (NCR3LG1), commonly called B7-H6, is a member of the B7 family of immune regulatory ligands. It is selectively expressed on the surface of various tumor cells and absent or very low in normal tissues. B7-H6 binds to the activating receptor NKp30 (NCR3) on natural killer (NK) cells, triggering NK cell activation, cytokine production, and direct cytotoxicity. Both membrane-bound and soluble forms of B7-H6 exist: the soluble form can impair NK cell function by masking NKp30 and preventing effective tumor cell killing. High levels of B7-H6 (especially the soluble form) are associated with worse prognosis in several cancers and may contribute to tumor immune evasion. Downregulation or blockade of B7-H6 sensitizes tumor cells to NK-mediated lysis, supporting its consideration as a biomarker, prognostic marker, and emerging immunotherapeutic target in oncology[1][3].
