Overview

Natural killer (NK) cell ligands are a heterogeneous group of cell surface molecules that regulate the cytotoxic activity of NK cells by binding to various activating and inhibitory receptors. In malignant or infected cells, the expression profile of these ligands is often altered; for instance, 'stress-induced' activating ligands such as MICA, MICB, and ULBPs are upregulated to signal distress to the immune system via the NKG2D receptor (Shimasaki et al., 2020, Nature Reviews Drug Discovery). Conversely, many tumors downregulate MHC class I molecules to evade T-cell recognition, a phenomenon known as 'missing-self' that normally triggers NK cell activation, or they overexpress inhibitory ligands like HLA-E to suppress NK cell function through the NKG2A receptor (Vivier et al., 2012, Science). Therapeutic strategies targeting these ligands include monoclonal antibodies that block inhibitory checkpoints, bispecific innate cell engagers (BiKEs) that bridge NK cells to tumor ligands, and CAR-NK cells engineered to recognize specific stress ligands (Long et al., 2013, Annual Review of Immunology).
