Natural killer cell p44-related ligand (NKp44L) refers to a group of molecules, most notably a specific isoform of the Mixed-Lineage Leukemia protein 5 (MLL5/KMT2E), that are expressed on the surface of tumor cells and stressed tissues but are absent in healthy cells (PMID: 23543761). These ligands serve as the primary binding partners for NKp44 (NCR2), an activating receptor found on Natural Killer (NK) cells and innate lymphoid cells (ILC3s). In the context of rheumatoid arthritis, NKp44L is upregulated on stressed synovial fibroblasts, where its interaction with NKp44-positive cells promotes the secretion of pro-inflammatory cytokines like TNF-alpha and IFN-gamma, driving chronic joint inflammation (PMID: 35154145). Beyond MLL5, other molecules such as BAG6, Nidogen-1, and PDGF-DD have been identified as NKp44 ligands, though their roles vary from activation to inhibition (PMID: 31434696). Therapeutically, NKp44L is an attractive target for precision medicine due to its restricted expression on pathological cells. Current research focuses on developing monoclonal antibodies to block the NKp44-NKp44L axis in autoimmune diseases or utilizing the ligand as a target for CAR-NK and CAR-T cell therapies in oncology (PMID: 34168115). However, the complexity of the NKp44 signaling network, which includes inhibitory ligands like PCNA, presents a challenge for drug development (PMID: 21441498). Safety concerns primarily involve the potential for off-target effects if the ligand is expressed on non-malignant cells undergoing physiological stress, as well as the risk of systemic cytokine release during potent immune activation.
Binding to the NKp44 (NCR2) receptor on NK cells to modulate cytotoxic activity and cytokine release; therapeutic strategies involve blocking this interaction or using the ligand as a target for cell-directed therapy.
