Overview

Natural killer cell p46 ligands (NKp46L) are a diverse group of molecules expressed on the surface of target cells, such as multiple myeloma cells, that serve as the primary recognition signal for the NKp46 (NCR1) activating receptor on Natural Killer (NK) cells (El-Sherbiny et al., 2007). The interaction between NKp46 and its ligands is a critical step in the innate immune system's surveillance and destruction of malignant plasma cells (Pazina et al., 2017). While the specific molecular identity of these ligands can vary, they include heparan sulfate proteoglycans and certain stress-induced surface proteins like vimentin (Hecht et al., 2009). In multiple myeloma, the expression levels of NKp46 ligands are often downregulated as the disease progresses, representing a key mechanism of immune evasion (El-Sherbiny et al., 2007). Therapeutic approaches aim to reverse this evasion by using immunomodulatory drugs (IMiDs) or histone deacetylase (HDAC) inhibitors to upregulate ligand expression and sensitize myeloma cells to NK cell attack (Jarahian et al., 2011). Consequently, NKp46 ligands are significant targets for both diagnostic assessment of immune susceptibility and therapeutic modulation in plasma cell dyscrasias.
