Overview

CD16 (FcγRIII) is a key surface receptor expressed on NK cells, neutrophils, monocytes, and macrophages. It mediates antibody-dependent cellular cytotoxicity (ADCC) by binding to the Fc portion of IgG antibodies, leading to the activation of immune effector cells and the elimination of antibody-coated target cells. There are two main isoforms: CD16a (FcγRIIIA), a transmembrane protein, and CD16b (FcγRIIIB), a GPI-anchored protein. CD16 is a crucial target in cancer immunotherapy strategies that enhance ADCC for improved tumor clearance. Its surface expression is regulated post-activation.
