Natural killer (NK) cell receptor-mediated cytotoxicity is the process by which NK cells recognize and eliminate target cells—such as virally infected or tumorigenic cells—through specialized surface receptors. This mechanism is a critical component of the innate immune response, providing rapid defense against abnormal cells without prior sensitization. It involves a balance of activating and inhibitory receptor signals, culminating in the release of cytotoxic granules or the induction of apoptosis in the target cell.
Target cell recognition via activating receptors (e.g., NCRs, CD16, TLRs), followed by granule exocytosis (perforin/granzymes) and/or death receptor (FasL/TRAIL) engagement, leading to target cell apoptosis or lysis.
