Overview

Natural killer cell receptor NKG2-D (NKG2D) is a type II transmembrane receptor predominantly expressed on natural killer (NK) cells and certain T lymphocyte subsets, including CD8+ T cells, γδ T cells, and NKT cells[1][3][5][6][8]. It is encoded by the KLRK1 gene and belongs to the C-type lectin-like family of immune receptors[1]. NKG2D functions as a potent activating and costimulatory receptor involved in the detection and elimination of stressed, transformed (tumor), or infected cells by recognizing a range of cellular stress-inducible ligands (notably MICA, MICB, and ULBP/RAET1 proteins)[1][3][6][8]. Upon ligand engagement, NKG2D triggers activation signals via adaptor protein DAP10 in humans (DAP10 and DAP12 in mouse isoforms), ultimately promoting immune cell cytotoxicity, cytokine production, and immune surveillance[1][2][3][5][6]. NKG2D is implicated in the pathogenesis and immune control of cancer, infections, autoimmune diseases, and transplant outcomes, making it a significant therapeutic and diagnostic target in immuno-oncology and immunotherapy research[3][5][6].
