NKp46 (NCR1) is a major activating receptor expressed on natural killer (NK) cells and some non-NK innate lymphoid cells. It recognizes ligands on stressed, infected, or transformed cells, triggering NK cell cytotoxicity and cytokine production. NKp46 plays a critical role in antiviral and antitumor immunity and is explored as a therapeutic target using bispecific antibodies and NK cell engagers.
Engagement of NKp46 leads to activation signals through associated adaptor proteins (FcεRIγ and CD3ζ), resulting in degranulation and cytokine production. Bispecific antibodies and NK cell engagers facilitate NK cell activation and target cell killing via NKp46 engagement.
