Overview

NKG2A is an inhibitory checkpoint receptor expressed mainly on NK and some T cell populations. It forms a heterodimeric complex with CD94 that recognizes HLA‑E ligands. Engagement transmits potent inhibitory signals through ITIM motifs—dampening cellular cytotoxicity—which plays critical roles in regulating immunity against tumors, infections, autoimmunity, inflammation, parasite infection, transplant rejection—and represents an emerging target for immunotherapeutic intervention.

Mechanism of action

Blocking antibodies against NKG2A prevent its interaction with HLA-E, relieving the inhibitory signal and enhancing NK/T cell cytotoxic activity.
