Natural killer group 2 member A (NKG2A) is an inhibitory receptor expressed primarily on natural killer (NK) cells and a subset of CD8+ T cells. It forms a heterodimer with CD94 and binds to HLA-E, delivering inhibitory signals that suppress activation, cytokine production, and cytotoxicity. NKG2A plays a crucial role in regulating immune responses, preventing excessive tissue damage and maintaining self-tolerance. It is a promising target for immunotherapy, particularly in cancer, where tumor cells often upregulate HLA-E to evade immune destruction. Blocking antibodies targeting NKG2A are under investigation to restore anti-tumor immunity.
