The Natural killer T cell receptor is a specialized antigen receptor expressed on NKT cells, a unique subset of T lymphocytes that share properties with both T cells and natural killer (NK) cells. This receptor is characterized by a semi-invariant αβ TCR—composed of an invariant alpha chain (Vα24-Jα18 in humans) paired predominantly with Vβ11—and specifically recognizes glycolipid antigens presented by the non-classical MHC class I-like molecule CD1d. Upon antigen binding, this receptor rapidly triggers NKT cell activation, leading to fast and robust cytokine release, modulation of immune responses, and direct cytotoxic activity. The NKT cell receptor plays an important role in cancer immunity, infection, autoimmunity, and immune regulation. Drugs like alpha-Galactosylceramide (α-GalCer) and related glycolipids interact with this receptor to harness NKT cell action, but therapeutic targeting faces challenges such as induction of anergy and risk of off-target immune effects. Its recognition and functional profile make it both a promising therapeutic target and a complex player in immunology[7][9][10].
