Overview

Natural moisturizing factor (NMF) is a complex collection of low-molecular-weight, water-soluble, hygroscopic substances found exclusively within the corneocytes of the stratum corneum (StatPearls, 2023). It is primarily composed of amino acids and their derivatives, such as pyrrolidone carboxylic acid (PCA) and urocanic acid, which are generated through the proteolytic degradation of the protein filaggrin (Journal of Investigative Dermatology, 2008). Other essential components include lactate, urea, and various inorganic ions like potassium and magnesium (Clinical, Cosmetic and Investigational Dermatology, 2013). NMF plays a critical role in maintaining skin hydration by binding water from the atmosphere and deeper epidermal layers, which ensures skin elasticity and facilitates the activity of hydrolytic enzymes required for desquamation (Rawlings & Harding, 2004). Deficiencies in NMF are strongly associated with skin barrier disorders, including atopic dermatitis and ichthyosis vulgaris, where reduced levels lead to xerosis, inflammation, and increased sensitivity to environmental irritants (Brown & McLean, 2012). Therapeutic strategies typically involve the topical application of NMF-mimicking substances or humectants to restore osmotic balance and improve barrier integrity (Journal of Clinical and Aesthetic Dermatology, 2012).
