Overview

Natural Moisturizing Factors (NMF) represent a complex collection of low-molecular-weight, water-soluble, hygroscopic substances found exclusively within the corneocytes of the stratum corneum. These factors primarily originate from the proteolysis of the protein filaggrin and include amino acids, pyrrolidone carboxylic acid (PCA), lactate, urea, and various electrolytes. Their primary biological role is to maintain adequate skin hydration by binding atmospheric water and retaining moisture within the epidermal layers, which is essential for skin elasticity and the proper functioning of desquamatory enzymes. A deficiency in NMF is strongly associated with various dermatological conditions characterized by barrier dysfunction, most notably atopic dermatitis and ichthyosis vulgaris, where reduced levels lead to xerosis (dry skin) and increased susceptibility to irritants. In the pharmaceutical and cosmetic industries, NMF is a major therapeutic target for topical agents designed to restore the skin's moisture balance. Drugs and emollients containing components like urea and alpha-hydroxy acids work by mimicking or replenishing these endogenous factors to improve the skin's physical properties and barrier defense.
