Overview

NAXD Antisense RNA 1 (NAXD-AS1) is a non-coding RNA transcribed from the antisense strand of the NAXD gene locus. It belongs to the long non-coding RNA class and is categorized as a natural antisense transcript. Antisense RNAs like NAXD-AS1 are increasingly recognized as important regulators of gene expression, often by overlapping with coding gene transcripts and influencing transcription, splicing, or chromatin state[1][3][4]. NAXD-AS1 has been catalogued genomically but is not well-characterized regarding its unique cellular function, protein interactions, or disease mechanism[3]. There is no specific evidence for drug interaction, biomarker utility, or direct clinical disease association beyond the mechanistic link to its sense gene, NAXD, whose deficiency is causative for rare neurological disorders[3]. The exact biological role of NAXD-AS1 remains to be elucidated.

Biological functions

Gene expression regulation (by antisense transcription)Possible chromatin modificationPotential involvement in alternative splicing and RNA stability (general for antisense RNAs)

Disease associations

Associated with NADHX dehydratase deficiency (via NAXD sense gene)Associated with progressive early-onset encephalopathy with brain edema and/or leukoencephalopathy, 2No NAXD-AS1–specific disease association has been mechanistically established; these are linked to the sense gene, not specifically the antisense transcript
