Nck proteins are adaptor molecules crucial for intracellular signal transduction. They consist of Nck1 and Nck2, which share high sequence similarity. Nck proteins are composed of three SH3 domains and one SH2 domain, facilitating protein-protein interactions that regulate actin cytoskeleton remodeling, cell migration, immune cell activation, and gene expression. They are primarily cytoplasmic and play a key role in linking receptor tyrosine kinases to downstream effectors. Dysregulation of Nck function is associated with diseases such as Wiskott-Aldrich Syndrome and tumor invasiveness.
