Nck-associated protein 5-like (NCKAP5L) is a protein involved in the regulation, organization, and stabilization of microtubules. It promotes microtubule growth and bundle formation by stabilizing microtubules, partially through increasing their acetylation. NCKAP5L localizes to the centrosome and microtubule plus-ends, and requires both tubulin binding and homodimerization for its role in microtubule bundle formation. Recent studies suggest that NCKAP5L is upregulated in multiple cancer types, especially colorectal cancer, where high expression correlates with patient survival and may serve as a promising diagnostic and prognostic biomarker. Genetic studies also associate NCKAP5L with autism spectrum disorder, although functional mechanisms are less well understood. Currently, there are no therapeutic drugs or established mechanisms of action targeting NCKAP5L directly, nor are there well-documented safety concerns related to its modulation[1][2][4][5][9][11].
