Overview

NCK interacting protein with SH3 domain (NCKIPSD), also known as SPIN90, is a multifunctional adaptor protein orchestrating the regulation of the actin cytoskeleton. It acts as a binding partner for various signaling molecules (including NCK, betaPIX, WASP, and IRSp53), promoting actin polymerization via the Arp2/3 complex and interaction with formin proteins. Through its SH3 and proline‐rich domains, it couples extracellular signals (such as PDGF and Src kinase) to local actin nucleation, facilitating processes like lamellipodia and stress fiber formation. NCKIPSD is essential for proper vesicle trafficking (e.g., EGFR turnover), plays a key role in cell shape, motility, dendritic spine plasticity, and has been linked to therapy-related leukemia due to chromosomal translocations, as well as cancer progression via its effects on cell mechanics and intracellular transport
