NCK1 antisense RNA 1 (NCK1-AS1), also known as the NCK1 divergent transcript (NCK1-DT) and SLC35G2-AS1, is a long non-coding RNA (lncRNA) encoded on chromosome 3q22.3. It does not code for a protein. NCK1-AS1 is implicated in regulating gene expression, particularly as a competitive endogenous RNA (ceRNA) that modulates various microRNAs (such as miR-133b, miR-137, miR-22-3p, miR-526b-5p, miR-512-5p, miR-138-2-3p, miR-6857) and downstream genes. It is associated with the pathogenesis and progression of several diseases, most notably multiple cancer types (e.g., gastric, lung, glioma, prostate, cervical cancers), where it often functions as an oncogene. NCK1-DT also appears to play a role in diabetes—its expression is differentially regulated in skeletal muscle and kidney tissues of diabetic patients, suggesting a tissue-specific function that may contribute to disease mechanisms or serve as a biomarker. Additionally, some reports propose potential roles in atherosclerosis. There are currently no known drugs that directly target NCK1-AS1/NCK1-DT, and it is not classified as a therapeutic drug target but may hold future value as a biomarker or indirect therapeutic target in oncology or metabolic diseases.
