Necator americanus aspartic protease 1 (Na-APR-1) is a 45 kDa cathepsin D-like aspartic protease expressed by the human hookworm Necator americanus, the most common cause of human hookworm infection worldwide[1][5]. Na-APR-1 initiates the proteolytic cascade that digests host hemoglobin in the parasite gut, a key process for parasite nutrition and survival[1][5]. It is a leading antigen candidate for hookworm vaccine development because neutralization of Na-APR-1 disrupts the parasite's feeding mechanism and reduces infection severity[1]. Na-APR-1 has been expressed and purified in heterologous systems for use in vaccine studies, with mutations eliminating hemoglobinase activity when used as an immunogen[1]. While no approved drugs directly target Na-APR-1, its essential biological role positions it as a molecular target for both vaccination and small-molecule inhibition strategies[1][5][3].
