Overview

Necator americanus glutathione S-transferase-1 is a nematode-specific enzyme (nu-class GST) involved in the detoxification of heme and metabolism of electrophiles, critical for the survival and blood-feeding of the adult human hookworm parasite. It has a unique heme-binding site and limited substrate range outside heme-related molecules, distinguishing it structurally and functionally from other GST families. Na-GST-1 is the lead antigen in ongoing vaccine development for hookworm infection, eliciting protective antibody responses and showing cross-species conservation that may provide protection against multiple hookworm species. Recombinant forms are produced in yeast for large-scale vaccine trials, and recent mRNA vaccine candidates further exploit its immunogenic properties.
