Overview

Necrotic DNA in the tumor microenvironment refers to fragments of nuclear and mitochondrial DNA released from tumor cells undergoing necrosis. This DNA acts as a damage-associated molecular pattern (DAMP), triggering innate immune responses and influencing the tumor microenvironment. While it can initially promote anti-tumor immunity, persistent exposure often leads to chronic inflammation and tumor progression by supporting angiogenesis, stromal remodeling, and recruitment of immunosuppressive cells.

Mechanism of action

Targeting DAMP signaling pathways (e.g., STING activation) may modulate immune responses against tumors. Understanding how released nucleic acids shape the microenvironment could inform strategies for immunotherapy or combination treatments targeting both cell death mechanisms and downstream inflammatory effects.
