Overview

Necrotic enteritis toxin B (NetB) is a potent pore-forming exotoxin produced by specific virulent strains of Clostridium perfringens, primarily identified as the essential causative agent of necrotic enteritis in poultry (Keyburn et al., 2008, PLoS Pathogens). As a member of the alpha-hemolysin family of beta-barrel pore-forming toxins, NetB monomers bind to the surface of host intestinal epithelial cells and oligomerize to form heptameric pores (Savva et al., 2013, Nature Communications). These pores lead to uncontrolled ion flux, cellular swelling, and eventual osmotic lysis, resulting in the characteristic necrotic lesions in the avian gut (UniProt Q2L9R5). Because NetB is critical for pathogenesis, it serves as a primary target for veterinary vaccines, including toxoids and recombinant protein formulations, aimed at inducing neutralizing antibodies. Research into NetB inhibitors and monoclonal antibodies continues to be a priority for the poultry industry to mitigate the significant economic impact of clostridial outbreaks.
