Overview

Necrotic tissue proteins represent a heterogeneous collection of denatured extracellular matrix components, intracellular proteins, and debris resulting from unregulated cell death (necrosis). In the context of wound management, these proteins form a physical barrier known as eschar or slough, which prevents the formation of healthy granulation tissue and serves as a medium for bacterial growth. While not a single molecular entity, necrotic tissue proteins serve as the therapeutic substrate for enzymatic debriding agents. These drugs, such as collagenase or bromelain-derived enzymes, selectively or non-selectively hydrolyze the peptide bonds within the necrotic mass to liquefy devitalized tissue. This process, known as enzymatic debridement, is essential for preparing the wound bed, reducing inflammation, and accelerating the transition to the proliferative phase of healing.
