Overview

Nectin-4 is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily and the nectin family of cell adhesion molecules. It mediates Ca²⁺–independent cell-cell adhesion and plays a key role in forming adherens junctions (AJs) and tight junctions (TJs). Nectin-4 is involved in various biological processes, including cell-cell aggregation, migration, proliferation of tumor cells, and EMT. It is overexpressed in multiple cancers and mutations cause ectodermal dysplasia-syndactyly syndrome type 1.

Nectin-4 is targeted in cancer therapy, particularly urothelial carcinoma, with the goal of disrupting cell-cell adhesion, proliferation, and survival signaling pathways.
