NEDD4-binding protein 2 (N4BP2) is a protein containing a polynucleotide kinase (PNK) domain near its N-terminus and a Small MutS Related (Smr) domain at the C-terminus[1][3]. The protein has enzymatic activities including 5'-polynucleotide kinase and nicking endonuclease activity, and binds ATP. It interacts with BCL-3 (an oncoprotein) and NEDD4, and may function as a substrate for ubiquitylation. N4BP2 likely plays a role in transcription-coupled DNA repair or genetic recombination. Structural studies indicate the Smr domain binds DNA and supports multimerization for DNA processing. Genetic variants in N4BP2 have been associated with increased risk of cancers, such as nasopharyngeal carcinoma, and developmental conditions like cleft lip and palate[1][3][5]. There is currently no evidence that N4BP2 serves as a direct drug target, nor are there known drugs or mechanisms of action targeting this protein specifically[3][1].
