NEDD4-binding protein 2-like 1 (N4BP2L1) is a cytoplasmic protein encoded by the N4BP2L1 gene, located on chromosome 13q13.1. It is ~243 amino acids in length and is involved in adipocyte homeostasis and glucose metabolism. N4BP2L1 is regulated transcriptionally by insulin/FoxO1 and USF1 and is required for GLUT4-mediated glucose uptake by interacting with the dynactin–dynein complex. High N4BP2L1 expression has been linked to poorer prognosis and metastasis in several cancers, while lower expression is noted in various breast cancer subtypes, suggesting a potential, yet unvalidated, use as a cancer biomarker[2][3][5]. No approved drugs are documented to act on N4BP2L1, and it is not currently considered a direct therapeutic target[2][5].
