NEDD4-binding protein 2-like 2 (N4BP2L2) is a nuclear protein acting as a transcriptional corepressor, implicated in the regulation of gene expression, particularly those related to hematopoietic stem cell differentiation and neutrophil maturation[1][2][4][5][6]. N4BP2L2 (also known as PFAAP5, or phosphonoformate immuno-associated protein 5) interacts with key regulators such as Gfi1 and neutrophil elastase, forming part of a feedback circuit essential for proper granulopoiesis[1]. Functional studies indicate that N4BP2L2 is vital for the repression of specific genes involved in neutrophil development; its silencing leads to impaired myeloid differentiation and can abolish neutrophil production, emphasizing its importance in maintaining blood cell homeostasis[1]. Although it is not currently recognized as a direct therapeutic target, mutations or dysregulation of N4BP2L2 have roles in inherited neutropenia syndromes, suggesting its indirect clinical relevance in hematological diseases[1][4][5]. No drugs are known to target N4BP2L2 directly, and it is not currently used as a biomarker or associated with specific therapeutic safety concerns.
