Nedd4 family interacting protein 1 (NDFIP1)

Nedd4 family interacting protein 1 (NDFIP1) is an evolutionarily conserved integral Golgi membrane protein characterized by three transmembrane domains and serves as an adaptor for the Nedd4 family of E3 ubiquitin-protein ligases[1][3]. It plays a critical role in regulating the ubiquitination and turnover of various substrates by recruiting Nedd4 family ligases such as NEDD4, ITCH, and NEDD4L, thereby influencing protein stability, vesicle trafficking, immune cell activation, and inflammation[3][4]. NDFIP1 helps maintain immune tolerance by promoting the degradation of specific signaling proteins in T cells and preventing chronic inflammation, as well as modulating pancreatic beta cell survival and cytokine responses. Pathologically, it has been implicated in breast cancer, immune disorders, and neurological conditions, but is not directly targeted by therapeutic drugs at this time[3][2].
