Neighbor of BRCA1 gene 1 protein (NBR1) is a highly conserved, ubiquitin-binding selective autophagy receptor that recognizes and directs ubiquitylated cellular cargo for macroautophagic degradation. It contains several protein interaction modules, including PB1, ZZ-type zinc finger, LIR (LC3-interacting region), UBA (ubiquitin-associated), four tryptophan (FW) repeat domains, and coiled-coil domains that coordinate cargo selection and autophagosome recruitment, often in concert with the related protein p62. NBR1 mediates the clearance of protein aggregates (aggrephagy), peroxisomes (pexophagy), and other organelles or structures such as focal adhesions and midbody remnants. In cancer, aberrant accumulation or overexpression of NBR1 can enhance tumor metastasis and immune evasion. NBR1 also plays important roles in plant and fungal stress responses. Aside from its role in autophagy, NBR1 is involved in cellular signaling and may have additional autophagy-independent functions[1][2][3][4].
