Overview

Neisseria meningitidis capsular polysaccharides from serogroups A, C, W/W135, and Y are key virulence factors and major antigens used in quadrivalent conjugate vaccines. The capsule protects the bacteria from host immune responses, preventing phagocytosis and complement-mediated killing. These polysaccharides are composed of distinct repeating units: O-acetylated N-acetyl-D-mannosamine phosphate for serogroup A, polysialic acid for serogroup C, sialic acid-galactose for serogroup W, and sialic acid-glucose for serogroup Y. The genetic determinants for capsule biosynthesis reside within the cps locus. These polysaccharides are targets for vaccines and important markers for epidemiological surveillance.
