The Neisseria meningitidis serogroup A capsular oligosaccharide is a carbohydrate antigen that constitutes the immunodominant portion of the serogroup A capsule of Neisseria meningitidis, a major cause of epidemic meningitis. The oligosaccharide, built from repeating units of O-acetylated (mainly at C3 and C4) N-acetylmannosamine-1-phosphate, forms a polysaccharide chain that coats the bacteria, enabling immune evasion. Vaccination induces antibodies against this oligosaccharide, conferring protective immunity. The epitope recognized by protective antibodies is found in fragments of 3–6 repeating units, and the presence and type of acetylation significantly impact antibody binding and bactericidal activity[1]. This target is widely used in conjugate vaccines where it is chemically linked to a carrier protein to enhance immunogenicity, particularly in infants and young children, thereby overcoming limitations of pure polysaccharide vaccines in these populations[1][3]. While no direct small-molecule drugs target this oligosaccharide, all serogroup A polysaccharide-conjugate vaccines rely on immune recognition of this structure. Safety concerns for this antigen are mainly related to the poor immunogenicity of the unconjugated polysaccharide in some age groups, necessitating conjugation to proteins in modern vaccines[3].
