Neisseria meningitidis serogroup B outer membrane vesicle (OMV) is a non-replicative, spherical nanoparticle shed by the outer membrane of Neisseria meningitidis serogroup B bacteria. It contains a complex mixture of outer membrane proteins (such as PorB, Opa, fHbp, NHBA, and others), lipids, lipopolysaccharide (LPS), and periplasmic components, and acts as a potent immunogen and adjuvant. OMVs are the basis of several vaccines (notably Bexsero/4CMenB), providing protection against meningococcal disease and additional cross-protection against Neisseria gonorrhoeae. Their immunogenicity derives from surface-exposed antigens and PAMPs, but their development as therapeutics requires measures to manage LPS-induced toxicity. OMVs are not classical molecular drug targets (like receptors or enzymes) but serve as multi-antigen complexes for vaccine, adjuvant, or immunomodulation purposes[2][3][4][5].
