Overview

The Neisseria meningitidis serogroup W-135 capsular polysaccharide is a surface-expressed carbohydrate capsule that is a principal virulence factor and major antigen defining the W-135 serogroup of N. meningitidis. It consists of alternating residues of sialic acid (N-acetylneuraminic acid) and D-galactose with α-(2→6) and α-(1→4) linkages; it is related to serogroup Y (which alternates sialic acid with D-glucose) but differs in sugar composition and O-acetylation pattern. The capsule protects meningococci against complement-mediated bactericidal activity and opsonization, facilitating bloodstream invasion and meningitis; antibodies to the capsular polysaccharide confer protective immunity. MenW-135 capsule is a component antigen in quadrivalent conjugate vaccines (ACWY), where conjugation to protein carriers enhances immunogenicity and induces functional bactericidal antibodies.
