Overview

Neocarzinostatin apoprotein is a 113-amino acid protein produced by Streptomyces carzinostaticus that functions as a specific carrier for the highly reactive neocarzinostatin chromophore (UniProt P02840). This apoprotein belongs to the enediyne-binding protein family and is essential for protecting the labile chromophore from nucleophilic attack and spontaneous degradation in aqueous environments (PubMed: 1551346). The resulting holo-protein complex, known as Neocarzinostatin or Zinostatin, is a potent antitumor antibiotic that targets DNA (DrugBank DB06159). The apoprotein facilitates the delivery of the chromophore to the minor groove of DNA, where the chromophore undergoes an activation process to generate a diradical species that causes double-strand breaks (PubMed: 2103035). While the apoprotein itself lacks cytotoxic activity, its high affinity for the chromophore is critical for the drug's stability and pharmacological profile (PubMed: 8401200). Clinical use of the complex has been explored for various malignancies, but challenges include its potential for immunogenicity as a foreign bacterial protein (PubMed: 3884400).
